Der JC-10-Assay ist eine empfindliche, fluoreszenzbasierte Methode zur Messung des mitochondrialen Membranpotentials , um Apoptose und Zellstress in lebenden Zellen zu überwachen. Im Vergleich zu JC-1 bietet JC-10 eine bessere Wasserlöslichkeit. Der Farbstoff akkumuliert in gesunden Mitochondrien (rot/orange) und diffundiert bei Apoptose als Monomer in das Cytosol (grün).

The Microbial Viability Assay Kit (WST Method) is a colorimetric, non-radioactive tool for rapidly quantifying viable microbes (bacteria/fungi) using water-soluble WST-8 salt. Metabolically active cells reduce WST-8 to an orange-colored formazan dye, with absorbance proportional to cell viability. It is commonly used for MIC (Minimum Inhibitory Concentration, e.g. antibiotics) determination.
Key Features and Principles:
Method: WST-8 is reduced to a soluble formazan dye by NADH/NADPH produced during microbial respiration (mediated by an intermediate electron acceptor).
Advantages: Unlike MTT, WST-8 produces a water-soluble dye, eliminating the need for organic solvent solubilization steps. It is less toxic to cells and allows for kinetic studies.
Application: Ideal for measuring bacterial cell viability, antibiotic susceptibility testing (MIC) against gram-positive and gram-negative bacteria, and antimicrobial screening.
Compatibility: Validated for various microorganisms including E. coli, S. aureus, and Candida species.
Limitations: Background absorbance can occur if samples contain high levels of reducing agents. The assay does not identify species, only total viable metabolic activity.
